The development of colorimetric techniques to measure the enzyme acetylcholinesterase inhibition by action of pesticides, is a relevant mechanism to identify and prevent poisoning with pesticides in risk populations. The objective of this research is to identify levels of toxic concentration of the organophosphate pesticides in human saliva compared the effects of these chemicals to the acetylcholinesterase enzyme through the modified Ellman method. Activity analysis in saliva samples of a group control of 25 non-exposed individuals was determined. The samples were treated and processed to maintain conditions of evaluation. The base used for the enzyme activity during the experiment was 3.326 U/L. From the various tests with different phosphorus compounds were obtained as for chemical structures having the enzymatic activity of acetylcholinesterase thion groups (parathion) using concentrations ranging 0-20 mg/L of the compound is 3.321 U/L. This results shows that pesticides which have an oxon group in its chemical structure, inhibits the acetylcholinesterase enzyme faster than those which have a thion group
