In vitro cytotoxicity evaluation: DNA fragmentation in murine thymocytes by organophosphate dimethoate

Abstract
Spanish title: Evaluación in vitro de la citotoxicidad y la fragmentación del ADN en timocitos murinos por efecto del organofosforado dimetoato. Los estudios sobre el efecto de los pesticidas en la salud son de gran interés debido a que pueden afectar diversos órganos o sistemas, tales como el sistema inmunológico, el cual es muy sensible a la exposición a contaminantes ambientales. Por tal motivo, el presente trabajo plantea el estudio de la evaluación in vitro de la citotoxicidad y la fragmentación del ADN en timocitos murinos los cuales fueron cultivados in vitro en diferentes concentraciones del organofosforado (0,05%, 0,1%, y 0,2%) y tiempos de 1, 3, y 6 horas respectivamente. Se evaluó el efecto sobre la citotoxicidad celular y el daño al ADN. Se concluye en base a los resultados obtenidos, que el dimetoato produce una citotoxicidad, generación de radical anión superóxido dependiente de la concentración y tiempo. Por otro lado, induce la generación de óxido nítrico en forma significativa para las concentraciones de 0,1% y 0,2%. En la evaluación electroforética de ADN, luego de 6 horas de incubación se observó fragmentación nucleosomal del ADN acompañada de un barrido, indicando una muerte celular por necrosis.
Studies on the effect of pesticides on health are of great interest because they can affect many organ systems such as the immune system which is very sensitive to exposure to environmental contaminants. Therefore, this paper presents the in vitro evaluation of the cytotoxicity and the DNA fragmentation in murine thymocytes , cultivated in vitro in different concentrations of organophosphate (0, 05 %, 0, 1 % and 0, 2 %) at intervals of 1, 3 and 6 hours respectively. The effect on the cell cytotoxicity, as well as the DNA damage was assessed. On the basis of the results obtained, it is concluded that dimethoate produces cytotoxicity, the generation of a superoxide anion radical which depends on the concentration and on the time. On the other hand it induces the generation of nitric oxide significantly for the concentrations: 0, 1 % and 0, 2 %. Electrophoretic evaluation of DNA after 6 hours of incubation showed a nucleosomal DNA fragmentation accompanied of a sweep, indicative of necrotic cell death.
