dc.description.abstractSamples from pond sediments were inoculated to obtain the pure culture of Methanosarcina sp. by repeated enriching and isolating. Growth was fasted on H_2+CO_2 or methanol. Colonies appeared in 1~2 weeks or in 3~4 weeks on acetate. They were usually about 1~2 mm in diameter and might be an irregular, angular shape when embedded in agar but circular in surface colonies. The color was usually, white to yellow. The characteristic pseudosarcina morphology made it easy identify Methanosarcina to genus level. Growth in liquid or solid medium produced methane and was greatly stimulated by addition of yeast extract. It fluoresced at 420nm.
